The 12 Principles of Green Chemistry

Anastas & Warner (1998) — "Green Chemistry: Theory and Practice"

1. PREVENTION
Better to prevent waste than to treat or clean up waste after formation.
Design reactions to produce no or minimal waste.

2. ATOM ECONOMY
Synthetic methods should maximize incorporation of starting
materials into final product. Design for 100% atom utilization.
   Atom Economy = MW(product)/Σ MW(reactants) × 100%

3. LESS HAZARDOUS SYNTHESIS
Use and generate substances with little or no toxicity
to human health and the environment.

4. DESIGNING SAFER CHEMICALS
Chemical products should preserve efficacy while
   minimizing toxicity. Benign by design.

5. SAFER SOLVENTS AND AUXILIARIES
Use of auxiliary substances (solvents, separating agents)
should be made unnecessary and, when used, innocuous.

6. DESIGN FOR ENERGY EFFICIENCY
   Energy requirements should be minimized.
Conduct reactions at ambient temperature and pressure when possible.

7. USE OF RENEWABLE FEEDSTOCKS
Raw materials should be renewable whenever technically and
   economically practicable. Bio-based > petroleum-based.

8. REDUCE DERIVATIVES
   Unnecessary derivatization (protection, temporary modification)
should be minimized — extra steps = extra waste.

9. CATALYSIS
Catalytic reagents are superior to stoichiometric reagents.
Catalysts: used in small quantities, increase selectivity.

10. DESIGN FOR DEGRADATION
Chemical products should be designed to break down into
    innocuous degradation products after use.

11. REAL-TIME ANALYSIS FOR POLLUTION PREVENTION
Analytical methodologies need to allow real-time monitoring
and control before formation of hazardous substances.

12. INHERENTLY SAFER CHEMISTRY
Choose substances and forms to minimize potential for
accidents (explosions, fires, releases to environment).

Mechanochemistry

Reactions without solvent using mechanical energy (grinding, milling):
  Ball milling: most common, scalable
  Twin-screw extrusion: continuous mechanochemistry
  Mortar and pestle: small scale

Advantages:
  No solvent needed (Principle 5)
  Often faster than solution reactions
Can access products not available in solution
Higher atom economy (no solvent waste)

Applications:
  Cocrystal formation (pharmaceuticals)
  MOF synthesis (metal-organic frameworks)
  Covalent bond formation (C-C coupling)
  Polymerization
  Pharmaceutical cocrystals, polymorphism control
  Nanoparticle synthesis

Examples:
Suzuki coupling: Pd-catalyzed C-C coupling by ball milling
Diels-Alder: solid state [4+2] without solvent
Knoevenagel condensation: K₂CO₃ catalyst, ball mill
Peptide coupling: solid-state amide bond formation

Common Pitfalls

PitfallFix
High yield = greenHigh yield + green metrics needed; stoichiometric reagents still waste
Bio-based = always betterLCA needed; some bio-processes have higher land/water use
Replacing solvent alone is enoughSolvents important but consider full process (energy, reagents, waste)
Atom economy ignores yieldUse RME (combines AE × yield × stoichiometric factor)
Catalysis always greenCatalyst synthesis, metal loading, recovery all matter
Mechanochemistry scales easilyScale-up of ball milling has engineering challenges

Best Practices

  • Prevent waste first — the most important principle (Principle 1)
  • Calculate metrics early — AE, E-factor, PMI during design phase
  • Consult solvent guides — CHEM21, GSK guides before choosing solvent
  • Consider the full LCA — don't just optimize one step
  • Design for end of life — biodegradability, recyclability from start
  • Use renewable energy — electrification of chemistry (electrochemistry)
  • Embrace biocatalysis — enzymes for chiral synthesis, mild conditions
